The PDH complex in cells that carry this mutation does not function. A series of experiments are performed to characterize the mutant PDH complex. When pyruvate coenzyme-A and NAD+ are added to purified PDH complex in vitro it is found that a very small amount of CO2 and acetyl-CoA are produced as the substrates are added and then the production of both of these compounds completely stops.
